Back in the late 2000s, 1992 Hosea Williams Drive was home to the quintessential Kirkwood bar, Aces Bar & Grill. Representative of the influx of young gentrifiers, the smoky dive bar served the local 20-somethings well. But times change and 20-somethings grow up. Enter The Pullman.

Partners and Kirkwood locals Heidi Shonkwiler and Dan Wingate set out to open a community-centric gastropub to match the evolving demographic of Kirkwood. When the pair learned that Aces was going under ?after a weeks-long stint as the kid-friendly Jak?s All In,? they saw their opportunity to build the pub the neighborhood needed. No longer an adults-only dive bar, The Pullman aims to offer a place to share a few pints with friends, as well as a place to park your stroller while you do it.

The Pullman walks that 30-something line between family-friendly enough to bring the kids along for dinner, but young enough to draw the crowd back in for drinks once the little ones are in bed. Wingate and Shonkwiler teamed up with chef Lotinza Clark from Radial Cafe to create a sampling of seasonal pub grub, offering a late-night menu most nights until midnight or later.

Unlike many self-declared gastropubs, shareable small plates are not the focus here, concentrating instead on sandwiches and entrees. The Oh George ($10), the house burger, comes with a man-sized half-pound patty of local beef, thick bacon slices, manchego cheese and a remarkably fresh yet mild jalapeno mayo. I?m pleasantly surprised by the special of the day: a fried chicken sandwich ($10) on toast, topped with tomato and mixed greens, oozing on all sides with melted pimento cheese.

Among the appetizers, a plate of the Sriracha wings ($8) makes for a great starter to split with friends. The sweet and spicy glazed wings keep just enough of the unmistakable kick of the rooster sauce, tempered just enough to satisfy heat lovers without alienating more timid palates. I was more than happy to hog most of our plate of chicken livers ($6), enjoying the light, flash-fried batter dusted with fresh herbs.

You?ll hear no complaints of small portions when the server airlifts the barbecue short ribs ($13) to your table. This impossibly large slab of slow-roasted beef arrives slathered with peach barbecue sauce atop a sweet potato and plantain hash, and can easily feed two adults. At seeing the shocked look in my eyes my server swears, almost apologetically, that I received the largest rib she?d seen them serve. This eats almost like a moist, braised brisket. Taken in human-sized portions, each bite is excellent, but I couldn?t help but picture some starving child across the world as I returned a plate with 2 pounds of meat still on it.

For a place claiming the gastropub title, I wish the beer selection was a little more extensive, currently offering 12 draft beers and a handful of bottles. However, half of the taps are dedicated to local breweries, and the rest offer a rotating selection of some more obscure beers that should keep most craft brew-heads interested.

Cocktails also get the seasonal treatment, and regularly change. The current menu offers multiple light libations, like the Palace ($8), a summery mix of gin, St. Germain and ginger ale, perfect for sipping on a warm afternoon. Or, spice things up a bit with the tequila-based Lightning Slinger ($8), mixed with pineapple juice, ginger liqueur and a dash of coarse ground black pepper.

A few missteps hint that the crew is still getting their feet under them . The blue cheese mayo is noticeably absent from my blackened chicken sandwich ($9) when I peel back the bun, and a bowl of spinach, artichoke and smoked gouda dip ($7) begs for a few more shakes of salt.

While the service is consistently attentive, I couldn?t help but notice the kitchen fall behind at seemingly odd times. One crowded Sunday brunch, I had no trouble getting my plate of peppered steak and eggs ($12), but on a slow Tuesday evening with only a handful of occupied tables, a plate of falafel and hummus ($7) arrives as we pay our check (for which our repentant server did not charge us.)

The Pullman aims to be the pub where all walks of the community can gather, and at that, they surely succeed. Shonkwiler and Wingate know their neighbors well, and have crafted a cozy little neighborhood tavern that everyone in the Kirkwood area should check out.

COLUMBUS, Ohio A new study has identified a life-and-death signaling role for a molecule on the surface of the immune cells involved in the most common form of chronic leukemia. The finding could lead to more effective therapy for chronic lymphocytic leukemia (CLL), an as yet incurable cancer that occurs in more than 16,000 Americans annually.

The study, led by researchers at the Ohio State University Comprehensive Cancer Center Arthur G. James Cancer Hospital and Richard J. Solove Research Institute (OSUCCC James), examines how an experimental drug called SMIP-016 kills CLL cells.

Earlier work by the same researchers showed that the drug targets a molecule called CD37, which is found on the surface of CLL cells. In this new study, the researchers discovered that the CD37 molecule has two regions that can concurrently activate two separate pathways in CLL cells, one that leads to cell death and another that promotes cell survival.

The findings show SMIP-016 activates the "death" part of the molecule, and they suggest that blocking the "survival" part of the molecule could improve the drug's effectiveness, the researchers say.

The study is published in the May issue of journal Cancer Cell.

"These findings open new possibilities for the use of immune-based therapy for treating CLL," says principal investigator Dr. John Byrd, a CLL specialist and professor of Medicine, of Medicinal Chemistry and of Veterinary Biosciences.

"We are already targeting the cell-death pathway using SMIP-016, and we might be able to optimize the drug's effectiveness by simultaneously blocking the cell-survival region of the molecule with a second antibody," he says.

Byrd notes that drugs that can block the cell-survival pathway already exist.

"We were very surprised and excited to find that CD37 is directly involved in regulating survival and cell-death pathways," says first author Rosa Lapalombella, a research scientist in Byrd's laboratory. "That's not usually the case for molecules of this kind."

Co-principal investigator Dr. Natarajan Muthusamy, associate professor of Medicine, notes that CD37 belongs to a family of molecules called tetraspanins, which usually only facilitate signaling by bringing together other molecules that when combined issue signals.

"This work is a great example of collaborative research involving several laboratories," says researcher and co-principal investigator Dr. Michael Freitas, associate professor of molecular virology, immunology and medical genetics, whose laboratory contributed to the global proteomic work of how CD37 signals. "The support for, and emphasis on, team science is a major driving strength of Ohio State and contributed to this project's findings," he says.

Byrd hopes to begin a clinical trial soon that tests SMIP-016 combined with a drug that blocks the survival pathway.

"Overall, these findings reinforce our belief SMIP-016 could be an effective agent for treating CLL and other malignancies that sometimes express the CD37 protein, including non-Hodgkin's lymphoma and acute lymphoblastic leukemia," says Byrd, who is the D. Warren Brown Chair of Leukemia Research.

Aaron Sorkin is an Academy and Emmy award winning writer and producer.

Hollywood screenwriter Aaron Sorkin was interviewed today by Walt Mossberg at the D10 conference in California. Sorkin has been hired by Sony to adapt Walter Isaacson?s official biography of Steve Jobs, and he has already tapped Apple co-founder Steve Wozniak to help advise him during the production process.

Sorkin talked about writing his Jobs movie today onstage. He described his writing process and how he wants to approach the film. After affirming that the film is still in the earliest stages of production, Sorkin talked about how he wants the audience to relate to his script and the kind of actor he wants to play Jobs.

To Sorkin, writing a script is a ?process of procrastination where you try to figure out what the movie is going to be about.? Echoing what he said earlier on the Jobs biopic, Sorkin remarked that ?it?s very difficult to shake the cradle-to-grave structure of a biography.? Instead, Sorkin wants to ?identify the point of friction that appeals to me and dramatize that.?

How long before Sorkin?s screenplay is ready for filming? ?I?m at the earliest possible stage,? Sorkin said. ?I?ll go through a long period that would not look to any causal observer like writing. It would look more like watching ESPN probably.? Sorkin was referring to the process of researching and gathering information on the late Apple co-founder.

?One of the hesitations I had in taking on the movie is that it was a little like writing about The Beatles,? said Sorkin. ?There are so many people out there who know so much about him and who revere him. I just saw a minefield of disappointment.? Sorkin has hope, however. ?Hopefully when I?m done with my research I?ll be in the same ballpark of knowledge about Steve Jobs.?

?Anytime you go to the movies and see ?the following is a true story?, you should think of it as a painting, not a photography,? said Sorkin. The Social Network, an Oscar-winning film on Facebook?s Mark Zuckerberg written by Sorkin, is the perfect example of what Sorkin refers to as ?taking an authorial view? of history. It?s a dramatization, not play-by-play snapshots of history.

When asked who would play Jobs in his movie with Sony, Sorkin said that the role had not yet been cast. ?They [the actor] are going to have to talk fast,? Sorkin said, referring to the intense way of speaking Jobs used to communicate. The actor will also have to be very smart, according to Sorkin. ?Actors can fake a lot of things, but intelligence is something you can?t fake.?

Ashton Kutcher, an actor known for his comedic roles in shows like That 70?s Show, is also starring in an indie Jobs biopic that will be out later this year, and Sorkin acknowledges the project by saying that Steve Jobs is a big enough persona for there to be ?room for more than one movie.?

?Steve Jobs is an extremely complicated guy,? said Sorkin. ?I know this for sure: I can?t judge the character.? Sorkin said that he has to ?find the parts of him [Jobs] that are like me? to defend the character.??With a character like Steve Jobs, you want to write the character as if they are making the case to God why they should be allowed into heaven.?

There were certainly rumors to the effect that a new iPad would have two ports, and as the seller notes there were actually leaked cases and drawings that showed this design. This particular iPad, however, is the first actual device to show up with the ports intact.

Whether Apple indeed intended to include both ports up until "the last minute" is anyone's guess, though it seems unlikely that Steve Jobs and Jonathan Ive, both decisive and minimalistic in their design tendencies, would allow such an ambiguity into a shipping product.

It is, however, possible and even likely that the decision of which side the dock would be on was left until the last minute, as the device is equally usable in both landscape and portrait modes. They settled?on portrait as the primary orientation, clearly, mirroring the iPhone and differentiating the iPad from widescreen monitors and TVs.

Interestingly, as a commenter points out at MacRumours, which found the listing, the circuitry necessary for the second dock connector is still present in normal iPads, though there is no hole in the case through which it can be accessed.

The real question, of course, is how did this device get out of Apple's labs in the first place? The seller tells Wired: "Judging by how Apple works, it?s most likely stolen, but I?m not sure about that...?I can?t give much information about the Apple device without revealing who specifically I am." That may put this auction in questionable legal territory.

Regardless, the package is already in the open, in transit?to the US-based buyer. Apple may try to track it down, but this cat is already out of the bag.

BEIRUT (AP) ? The U.N.'s human rights office said Tuesday that most of the 108 victims of a massacre in Syria last week were shot at close range, some of them women, children and entire families gunned down in their own homes.

The massacre on Friday in Houla drew new international outrage, with more than half a dozen countries including France and Britain expelling Syrian diplomats in protest in what the White House said was a sign of their "absolute disgust" with Assad's rule.

Still, spokesman Jay Carney said the White House remains opposed to military action in Syria, reasoning that would only lead to more carnage.

The U.N. peacekeeping chief Herve Ladsous also said there are strong suspicions that pro-government fighters were responsible for some of the killings.

"We are at a tipping point," special envoy Kofi Annan told reporters Tuesday in Damascus, following a meeting with President Bashar Assad. "The Syrian people do not want the future to be one of bloodshed and division."

He called on the government and the armed opposition to stop all violence.

The U.N. report indicated that most of the dead were killed execution-style, with fewer than 20 people cut down by regime shelling. The U.N. cited survivors and witnesses blaming the house-to-house killings on pro-government thugs known as shabiha, who often operate as hired muscle for the regime.

"What is very clear is this was an absolutely abominable event that took place in Houla, and at least a substantial part of it was summary executions of civilians, women and children," said Rupert Colville, spokesman for the U.N. High commissioner for Human Rights. "At this point, it looks like entire families were shot in their houses."

Houla activists reached by Skype said government troops shelled the area after anti-government protests on Friday and clashed with local rebels. Later, shabiha from nearby villages swept through the area, stabbing residents and shooting them at close range.

Videos posted online by anti-regime activists show explosions in Houla, dismembered bodies lying in the streets, then row upon row of the dead laid out before being buried in a mass grave. Some of the videos showed dozens of dead children, some with gaping wounds.

The Syrian regime has denied any role in the massacre, blaming the killings on "armed terrorists" who attacked army positions in the area and slaughtered innocent civilians. But it has provided no evidence to support its narrative nor has it given a death toll.

U.N. investigators have said they found tank and artillery shells in Houla after the attack, but stopped short of blaming regime forces for the killings.

The U.N. said that at least 108 people, including 34 women and 49 children, were killed in the attack that began on Friday and continued through the night on a group of poor farming villages northwest of the central city of Homs.

Speaking to reporters in Geneva, Colville said U.N. monitors who visited the area found that fewer than 20 of the dead were killed by artillery fire. The rest appeared to have been shot at close range.

He said information from U.N. investigators and other sources indicated that many of the victims were killed in the Houla village of Taldaw in two separate incidents. Local residents blamed the killings on pro-regime militias known as shabiha, which sometimes act "in concert" with government forces, he said.

He said a fuller investigation was needed before he could comment on that, and called on Syria to allow free access to U.N. investigators.

The brutality of the killings and the high death toll raised new questions about the ability of a U.N.-brokered plan to end the violence in Syria, which began in March 2011.

According to the state-run news agency, SANA, Assad blamed terrorists and weapons smugglers for scuttling Annan's peace plan. The regime denies there is any popular will behind the country's uprising, saying foreign extremists and terrorists are driving the unrest.

The new information provided by the U.N. draws attention to the role of the shabiha in 15 months of violence in Syria. Assad's government often deploys pro-regime thugs or armed militias to repress protests or carry out more military-style attacks on opposition areas.

They frequently work closely with soldiers and security forces, but the regime never acknowledges their existence, allowing it to deny responsibility for their actions.

A Syrian official denied again on Tuesday any involvement.

"It is irrational that any party who wants to make Annan's mission a success would ever commit such a massacre," Deputy Foreign Minister Faisal Mekdad told reporters. He said Syria remained committed to Annan's plan and "had not committed a single violation."

Activists have posted videos of tanks and armored vehicles in the middle of cities, a violation of the plan, and U.N. observers said they found spent tank and artillery shells in Houla after the massacre there. Funeral videos also showed local rebels among the mourners ? making it unlikely they carried out the killings.

Anti-regime rebels around the country regularly attack military convoys and checkpoints, killing soldiers.

Syria's international isolation deepened in response to the killings. Governments around the world expelled Syrian ambassadors and diplomats Tuesday, an unusual, coordinated blow to Assad's regime.

The United States, Britain, Canada, Australia, France, Germany, Italy, Spain and the Netherlands took action Tuesday against Syrian diplomats. Britain's foreign secretary said the countries involved in Tuesday's expulsions would also push for tougher sanctions against Syria.

Longtime Syrian ally Russia has largely stood by Damascus, although Moscow is growing increasingly critical ? particularly over the Houla massacre. On Tuesday, however, Russian Foreign Minister Sergey Lavrov accused unnamed countries of trying to use the Houla killings "as a pretext for taking military measures."

He said such nations sought to impede Annan's plan because it seeks dialogue between Syrians, not regime change.

Syria's unrest began in March 2011, with protests calling for political change. Government troops swiftly cracked down at the uprising spread, and many in the opposition have taken up arms to defend their towns and attack government troops.

ScienceDaily (May 29, 2012) ? Computer chips of a type more commonly found in games consoles have been used by scientists at the University of Bristol to reveal how the flu virus resists anti-flu drugs such as Relenza and Tamiflu.

Professor Adrian Mulholland and Dr Christopher Woods from Bristol's School of Chemistry, together with colleagues in Thailand, used graphics processing units (GPUs) to simulate the molecular processes that take place when these drugs are used to treat the H1N1-2009 strain of influenza -- commonly known as 'swine flu'.

Their results, published May 29 in Biochemistry, provide new insight that could lead to the development of the next generation of antiviral treatments for flu.

H1N1-2009 is a new, highly adaptive virus derived from different gene segments of swine, avian, and human influenza. Within a few months of its appearance in early 2009, the H1N1-2009 strain caused the first flu pandemic of the 21st-century.

The antiviral drugs Relenza and Tamiflu, which target the neuraminidase (NA) enzyme, successfully treated the infection but widespread use of these drugs has led to a series of mutations in NA that reduce the drugs' effectiveness.

Clinical studies indicate that the double mutant of swine flu NA known as IRHY2 reduced the effectiveness of Relenza by 21 times and Tamiflu by 12,374 times -- that is, to the point where it has become an ineffective treatment.

To understand why the effectiveness of Relenza and Tamiflu is so seriously reduced by the occurrence of this mutation, the researchers performed long-timescale molecular dynamics (MD) simulations using GPUs.

Professor Mulholland said: "Our simulations showed that IRHY became resistant to Tamiflu due to the loss of key hydrogen bonds between the drug and residues in a part of the NA's structure known as the '150-loop'.

"This allowed NA to change from a closed to an open conformation. Tamiflu binds weakly with the open conformation due to poor electrostatic interactions between the drug and the active site, thus rendering the drug ineffective."

These findings suggest that drug resistance could be overcome by increasing hydrogen bond interactions between NA inhibitors and residues in the 150-loop, with the aim of maintaining the closed conformation.

The research was supported by the Engineering and Physical Sciences Research Council (EPSRC) through a Leadership Fellowship grant to Professor Mulholland and a software development grant to Professor Mulholland and Dr Woods.

UN envoy discusses peace plan with Syrian leader

BEIRUT (AP) ? International envoy Kofi Annan is meeting with President Bashar Assad to discuss the peace plan meant to end the country's 15-month-old crisis.

That plan appears increasingly shaky following a massacre of more than 100 people, many of them women and children, last week. The U.N. said government forces fired tank shells and artillery at the villages that make up Houla, but stopped short of blaming them for the killings.

Activists said most of the dead were killed by pro-government thugs who stormed the area.

The Houla killings prompted sweeping international condemnation and the harshest language yet from Syrian ally Russia ? making it a potential turning point in the crisis that has killed more than 9,000 people.

The meeting between Annan and Assad is taking place Tuesday in Damascus.

BEIRUT (AP) ? A weekend massacre of more than 100 people emerged as a potential turning point in the Syrian crisis Monday, galvanizing even staunch ally Russia to take an unusually hard line against President Bashar Assad's government.

Analysts said Russia may be warning Assad that he needs to change course or lose Moscow's support, which has been a key layer of protection for the Syrian government during the uprising that began in March 2011.

Russia has grown increasingly critical of Damascus in recent months, but Foreign Minister Sergey Lavrov's latest comments were unusually strong. Although he said opposition forces have terrorists among them, he put the blame for 15 months of carnage primarily on Assad's government.

"The government bears the main responsibility for what is going on," Lavrov said in Moscow following a meeting with British Foreign Secretary William Hague. "Any government in any country bears responsibility for the security of its citizens."

Alexei Malashenko, a Middle East expert with the Carnegie Moscow Center, said Lavrov's comments suggest Russia may be backing away from its long-standing support for Damascus.

"Bashar Assad is driving himself and Russia into a corner," Malashenko said. "Bashar has definitely gotten the sense that he may lose Russia's sympathy, and he may step back a bit."

It is not clear whether Assad's forces were exclusively to blame for the slaughter of 108 people Friday in Houla, a collection of poor farming villages in Homs province. The United Nations said 49 children and 34 women were among the dead; some had bullet holes through their heads.

The U.N. Security Council blamed Syrian forces for artillery and tank shelling of residential areas, but it did not clearly state who was responsible for the close-range shooting deaths and "severe physical abuse" of civilians.

Activists from the area said the army pounded the villages with artillery and clashed with local rebels. They said pro-government gunmen later stormed the area, doing the bulk of the killing by gunning down men in the streets and stabbing women and children in their homes.

The Syrian government rejected that account entirely, saying soldiers were attacked in their bases and fought back in self-defense without leaving their bases.

Russia blamed both the government and the rebels for the Houla massacre.

"Both sides have obviously had a hand in the deaths of innocent people, including several dozen women and children," Lavrov said. "This area is controlled by the rebels, but it is also surrounded by the government troops."

He said Russia has no interest in propping up Assad but wants Syria to guide its own transition under a plan brokered by special envoy Kofi Annan.

"We don't support the Syrian government; we support Kofi Annan's plan," Lavrov said.

Moscow's pro-Syria stance has been motivated in part by its strategic and defense ties to Damascus, including weapons sales. Russia also rejects what it sees as a world order dominated by the U.S.

Losing Russian support could be disastrous for Assad because his crackdown has left him almost completely isolated internationally. Russia and China have stood by him so far, using their veto power to block U.N. resolutions against him.

Hilal Khashan, a political science professor at the American University of Beirut, said the Houla massacre appears to be ushering in a change in Russia's position.

"There is a shift and the momentum against the regime is gathering," Khashan said. "The momentum is building and the Russians are not blocking the rising momentum."

The Syrian conflict is among the most explosive of the Arab Spring, in part because of Syria's allegiances to powerful forces, including Lebanon's Hezbollah and Shiite powerhouse Iran.

Activists say as many as 12,000 people have been killed since the uprising began. The U.N. put the toll as of March, a year into the uprising, at 9,000, but many hundreds more have died since.

After 35 years of eavesdropping on the stars, hoping to hear the intelligent murmurings of an extraterrestrial civilization, the SETI Institute?s lead alien hunter is hanging up her receiver and retiring as director of the Center for SETI Research.

But she?s not quitting the quest.

Instead of querying the heavens for signs of life, Tarter will query life on Earth for the funds needed to keep Earth?s ears online. ?We?re just going to make this a crusade,? she told me during an interview. (You can read it here.)

Physicist Gerry Harp has taken the reins from Tarter, the SETI Institute announced on May 22. ?It?s been challenging and exciting, a lot of new things to learn, a lot of new meetings to go to,? Harp says of his new role. ?But from what I?m seeing, Jill?s working at least as hard as she ever did before. I think we should acknowledge that she is still very much a part of the program.?

Tarter?s curiosity about life on other worlds erupted in the 1970s, fueled by a serendipitous merging of her ability to program a very old computer (a PDP-8/S ? ?I always thought the ?S? stood for ?stupid,?? she says), with a forward-looking proposal to find out whether other communicating civilizations exist in the galaxy. At the time, Tarter was in graduate school at the University of California, Berkeley. ?I couldn?t imagine anything more exciting or rewarding than trying to answer this old question,? Tarter says. ?I got hooked and I stayed hooked.?

In 1984, Tarter and several colleagues founded the SETI Institute, a nonprofit organization that has since grown to include more than a hundred people. Once focused mainly on detecting radio signals from extraterrestrial civilizations, the institute now employs scientists studying many factors important for the existence of alien life ? from Kepler team members who are detecting and characterizing exoplanetary systems, to astrobiologists searching for life in the most extreme environments on Earth and in the solar system?s exotic nooks and crannies.

But the institute needs money to keep searching. Once funded by a fraction of a percentage of NASA?s budget, SETI has relied on private donors to maintain the interstellar dial tone since 1993 ? courtesy of one senator, who killed any substantial SETI funding for the foreseeable future. ?For years, we became the four-letter S-word that you couldn?t actually say at NASA headquarters,? Tarter recalls.

Now, with the Allen Telescope Array ? a SETI array comprising 42 dishes (out of a planned 350) ? that funding is more crucial than ever. Last April, the array went into hibernation after funds dried up. In October, the array returned to operations, propelled by individual donors who contributed more than $200,000. Now, the telescope is under new management by SRI International, and is supported by a contract from the U.S. Air Force that tasks it with searching for orbiting space trash.

Still, 50 percent of the observing time is occupied with SETI searches. ?For SETI, it is the best thing around,? Harp says. ?We have 12 hours a day on this telescope. It?s very difficult to get 12 hours a day on any telescope in the world, and that?s what we have.?

Tarter is hoping to once again tap the enthusiasm of SETI supporters and build a lifeline that will support the search for centuries. ?SETI really is the investment in our future,? she said. ?If we detect a signal, we know we can have a long lifetime.?
